Binding Conditions/Buffer

RPMI-1640 (pH 7.2) supplemented with BSA (1 mg/mL, Sigma, St Louis, MO), yeast tRNA (0.1 mg/mL, Invitrogen)

Assay

Binding isotherm: Putative aptamer sequences were labeled at the 5′ end with [32P]. Five million cells per well were incubated with increasing concentrations of labeled sequences in bufer. Afer three washes, the amount of 32P-labeled aptamer associated with cells was determined by liquid scintillation counting. Data were normalized by fraction bound. Malaria.1, Malaria.2 and Malaria.3 bind to IEs with high afnity but the randomly generated control sequence of the same length (RDM1) shows no binding to IEs
