Assay

Determination of binding affinities using fluorescence intensity. A constant concentration of protein was immobilized onto a 96-well plate and blocked with 5% BSA. Each well was incubated with a different concentration of aptamer (0–5000 nM). After several washing steps, the fluorescence intensity of the DNA was measured in each well surface and the corrected signal was normalized using the BSA-only control. Fluorescence intensities (in relative fluorescence units, RFU) were plotted against the aptamer concentration (in lM), and the binding affinity KD was calculated using nonlinear regression on GraphPad Prism 5. Experiments were performed in triplicate
